Teacher
After graduation, Vasudev Devnani began his teaching career and became a dean at Vidhya Bhawan Polytechnic College in Udaipur. He was the dean of the college for 12 years.
Politics
During college, Vasudev Devnani was highly influenced by the ideologies and practices of the Rashtriya Swayamsevak Sangh (RSS); therefore he joined the RSS. Later, he joined the student wing of RSS, i.e., Akhil Bharatiya Vidyarthi Parishad (ABVP). He served as the Rajasthan state president of Akhil Bharatiya Vidyarthi Parishad (ABVP) for nine years before he joined the Bharatiya Janata Party.

In 2003, Vasudev Devnani was selected to contest as a BJP candidate against Congress leader Naren Bhagat Shahani from the Ajmer North constituency in the Rajasthan assembly elections with more than 45 per cent of the total votes to his name and became the MLA of the Ajmer North constituency. In December 2003, in theĀ Vasundhara RajeĀ government, he was appointed as the Minister of State for Education. Vasudev Devnani won the Rajasthan assembly elections from the Ajmer North constituency in 2008 and again in 2013. In 2008, he won the elections by only 688 votes. In 2013, he was given an independent charge of primary and secondary education in the Ministry of Vasundhara Raje. In 2018, he again contested for the Ajmer North constituency against the Congress leader Mahendra Singh Ralawata and won by a margin of 8,630. For the fifth time consecutively, in 2023, Vasudev Devnani contested the state legislative assembly elections from the Ajmer North constituency against the Congress leader Mahendra Singh Ralawata and won with a margin of 4,644 votes.

On 12 December 2023, Vasudev Devnani was appointed as the 18th Speaker of the Rajasthan Legislative Assembly in the Bhajan Lal Sharma government of Bhajan Lal Sharma.
