She worked as an emcee at Asiant from 2006 to 2010. In 2011, she started working as an emcee in many shows on Amrita TV such as the game show Q-Matrix and the musical reality show Super Star-The Ultimate; she worked for Amrita TV until May 2012.

Santhi Mayadevi made her debut in the Malayalam film industry in 2019 with the film ‘Ganagandharvan,’ which featured Mammootty and Vanditha Manoharan in the lead roles; she played the role of Advocate Anamika in the film.

She later acted in many films including the Malayalam version of Drishyam 2 in 2021 in which she played the role of Advocate Renuka alongside Mohanlal and Anjali Nair in lead roles. In 2022, she played the role of SP Reena in the film ‘Naalam Mura.’
